From Sydney to Siberia, revelers prepare to celebrate the dawn of a new year.

Updated at 6:05 ET: Happy new year! At least to those on Christmas Island. The Australian territory rang in 2013 at 5 a.m. ET on Monday. So did Kiribati and Samoa. They were the first populated places to do so.

More populous areas, including New Zealand, flipped the calendar just an hour later, joined by Fiji and Tonga.

Meanwhile, Scotland was already a day into the party.?International datelines hadn't been surreptitiously changed. Scotland was celebrating the annual festival known as Hogmanay, which kicked off Sunday night with thousands of torchbearers marching in Edinburgh to bid farewell to 2012 in a celebration drawing inspiration from pagan traditions. The Scotsman newspaper estimated that 7,000 people participated in the "river of fire" through the city center.

The evening of celebration was just the start of a fete lasting until Wednesday and expected to draw 80,000 revelers from around the world, according to the official Hogmanay website.?

Thousands of miles to the east, the party is a little bigger. In Sydney, eager revelers camped Sunday night on the shores of the harbor -- the first wave of an estimated 1.5 million people expected to watch a fireworks display centered on the Sydney Harbor Bridge.?

Another 2 million Australians, out of a population of 22 million, were expected to watch the show?on television,?Sydney's Lord Mayor Clover Moore told reporters. ?Entertainment for millions doesn't come cheap -- the extravaganza headlined by pop singer?Kylie Minogue?carried a?price tag?of $6.9 million.?

In a field in Myanmar's largest city, Yangon, workers tested a giant digital countdown screen early on Monday against a backdrop of the revered Shwedagon pagoda.

The celebration was set to be the first public New Year's countdown in Myanmar, a country ruled for almost five decades by military regimes that discouraged or banned big public gatherings.

No. 2 UConn women rout No. 1 Stanford 61-35

STANFORD, Calif. (AP) ? Geno Auriemma insists long winning streaks do little to get him amped up, even after he traveled his Connecticut team across the country to face top-ranked Stanford and its daunting 82-game unbeaten run at Maples Pavilion.

The Huskies sure got a thrill from leaving with an unexpected rout and silencing a typically raucous crowd ? and likely stealing away the No. 1 spot in the polls along with it.

Kaleena Mosqueda-Lewis and second-ranked UConn played spoiler and streak-buster this time, snapping Stanford's nation-leading home winning streak with a surprisingly easy 61-35 victory Saturday.

It was the Huskies who saw the end of their NCAA record 90-game winning streak at Maples Pavilion with a 71-59 loss two years ago, almost to the day on Dec. 30.

"The last time we were here it was a big event for them and they treated it like it was a big event, and God bless them," Auriemma said. "But for us to win a game here in late December, it's no more than just a big game against a really good team. The other stuff, I didn't get all that excited about us winning 90, I'm not going to get that excited about us beating somebody who's won 82 in a row at home."

Mosqueda-Lewis scored 19 points as UConn (11-0) thoroughly outplayed Stanford (11-1) on both ends of the floor in this highly touted game featuring the country's top programs and Final Four regulars from opposite coasts.

Stanford got harassed right off the home floor that it ruled with perfection and dominant play for nearly six years ? and the Huskies surely took the Cardinal's No. 1 spot along with it.

The game was the 51st meeting between the top two teams in the poll. The No. 1 seed had won the previous nine matchups and holds a 31-20 lead.

Chiney Ogwumike had 18 points and 13 rebounds but struggled in the post as Stanford lost at home for the first time since March 2007.

"It was a bad day. Whatever we were trying to do, we really struggled with it," Stanford coach Tara VanDerveer said. "Connecticut came in here on a mission. Mission accomplished."

Stefanie Dolson had 10 points and 14 rebounds and flustered Ogwumike all afternoon, and Bria Hartley and Kelly Faris also scored 10 apiece for UConn.

Breanna Stewart scored seven straight points during a decisive 17-2 run in the first half as UConn built a 22-7 lead against the cold-shooting Cardinal and kept the pressure on the rest of the way.

Stanford trailed 31-13 at halftime in one of its worst 20 minutes in recent memory ? though the school had no record of when the Cardinal were last held to that few points in a half.

They had gone five full seasons with an unbeaten record on their home floor.

Mosqueda-Lewis, last season's Big East top freshman averaging 16.4 points and 5.0 rebounds, shot 7 for 13 as Connecticut shot 37.5 percent from the floor ? and that was plenty good enough against a Stanford team that wound up at a dismal 19.3 percent (11 for 57) in its lowest single-game shooting performance and fewest field goals made.

"I love the game, I think it's a great game for both teams, but for our game two years ago it was a national event ? I don't think tonight was a national event," Auriemma said. "We beat a really good Stanford team on their home court and they happened to have won a lot of games at home."

Stanford had four early possessions over the opening 4 1/2 minutes in which it faced a dwindling shot clock.

This wasn't the Cardinal's worst home loss ever ? that was a 96-51 defeat to Long Beach State on March 10, 1983 ? though UConn sure made it feel like it for an afternoon to silence a typically animated crowd.

Auriemma was booed by the crowd during pregame introductions, then again when the coach was whistled for a technical foul with his team ahead 19 points with 13:22 remaining.

UConn, which had trailed for only 48 seconds all season coming into the game and now just 2:21 in all, won for the first time at Stanford in four tries.

Ogwumike, Stanford's leading scorer and rebounder averaging 21.8 points and 12.8 boards, was held to 6-for-22 shooting.

"I don't think I've experienced anything like this," Ogwumike said. "That Connecticut team we played for 40 minutes is the standard. And the great thing is now we've experienced the standard."

UConn's 6-foot-5 Dolson made things tough all day for Ogwumike ? who stands 6-4 ? and gave her problems with a size advantage to deny Stanford's top player and keep her away from the basket. That led to some forced shots in the first half, when Ogwumike missed 8 of her first 9 attempts. She scored at the 7:25 mark of the first half to end a nearly 6-minute scoring drought since her previous basket on a putback at 13:19.

Stanford then went 4:58 without scoring before Ogwumike's three-point play 2:27 before halftime.

"Stefanie was going to guard Chiney until she proved that she couldn't," Auriemma said.

Dolson was whistled for her third foul with 17:42 left in the game, but Mikaela Ruef missed both free throws.

Walking off the court after this one "was complete opposite," Dolson said.

"Two years ago we lost and it ended our streak. It was just a horrible loss," she recalled.

Bonnie Samuelson's back-to-back 3-pointers midway through the second half, the Cardinal's first from behind the arc, gave them some life. But it was short-lived as the Huskies pulled away.

Stanford's 82-game home winning streak dated to a 68-61 loss to Florida State in the second round of the NCAA tournament on March 19, 2007.

This season's Cardinal stunned defending NCAA champion and then-top-ranked Baylor and Brittney Griner on Nov. 16 to become the nation's No. 1 team and avenge a loss in last spring's NCAA semifinals.

The Cardinal hardly looked like the best team Saturday against the dominant Huskies. They were held below 40 points for the first time since a 72-32 loss to Missouri on Jan. 2, 1984.

Stanford missed its initial 10 3-point tries and began the game 3 for 22 to fall behind 22-7, with UConn getting three quick 3s.

Ogwumike's big sister, Nnemkadi ? the 2012 No. 1 overall WNBA draft pick of the Los Angeles Sparks ? attended the game, along with other former Stanford players Rosalyn Gold-Onwude and Lindy La Rocque.

Stanford senior Joslyn Tinkle, the team's second-leading scorer, managed only two points while playing on her 22nd birthday.

Over the years, more than a few alternate reality games, transmedia storytelling projects, and advertising campaigns have warned that December 2012 would mark the end of the world. That shouldn?t come as much of a surprise: you can?t get much higher stakes than saving the world, and fighting against an ancient prophecy with its own pre-existing mythos (however misinformed) can add mystique to a narrative.

In 2009, the alternate reality game?The Institute for Human Continuity?reportedly sparked?hundreds of letters?to NASA?s Astrobiology Institute from people earnestly worried about the coming Apocalypse.?Closer to the date in question, Funcom?s The Secret World offered a series of six missions intended to forestall the End of Days both inside and outside the game. Even Old Spice got in on the harbinger of doom act, using points from six increasingly ridiculous flash games to power a laser cutter that slowly etched additional time onto the Mayan calendar for their absurdist campaign, Old Spice Saves the World.

Proclamations of impending disaster weren?t limited to global catastrophe this year, with Fourth Wall Studios? Elan Lee adding his voice to the chorus?claiming that ARGs are dead at the StoryWorld Conference in Los Angeles.?And yet, 2012 was in many ways a renaissance for alternate reality games and transmedia storytelling, as new sources of funding arise for a thriving community of developers. What follows is a closer look at some of the major events in alternate reality gaming for the year.

A Quick Look at the Industry

The most heartening development of the year is the growing influence of local groups of transmedia developers. Transmedia LA turned to Kickstarter to raise almost $10,000 to develop the alternate reality game Miracle Mile Paradox?to give its members hands-on experience creating a game from start to finish, calling back to the ?training ARG? Orbital Colony. In New York City, StoryCode?s focus was on a more condensed Story Hackathon. Inspired by Storycode?s hackathon, Transmedia SF ran its own Transmedia Jam. The hackathon model will be moving to Los Angeles in 2013, opening even more opportunities.

Established institutions in transmedia funding and support like?Power to the Pixel?and Sundance?s?New Frontier?are ramping up their efforts to provide direction and funding for the field, while new entrants like the?Tribeca Film Institute, the?National Endowment for the Humanities, and the?Canadian Film Centre?have been quick to follow with a variety of programs for prospective developments. On the legal front, Stitch Media?managed to prevail?in a Nova Scotia Supreme Court case revolving around the definition of interactivity for new media projects.

The list of transmedia storytelling platforms available for public consumption grew longer this year, as teams are increasingly viewing the technological infrastructures developed for projects as potential revenue generators. The Shadow Gang introduced Galahad, the platform developed in conjunction with Michael Grant?s alternate reality game for his Bzrk?book series, while Failbetter Games released its StoryNexus platform into the wild, after using it to produce games like?Echo Bazaar?(now known as Fallen London)?and The Night Circus. The year saw the official launch of Fourth Wall Studios? RIDES platform. The company recently laid off the majority of its staff?to shift the company?s focus away from production, redoubling efforts on developing the platform for licensing to partners.?These new platforms join existing alternatives like Social Samba and Conducttr as potential tools for content management.

Return of the Video Game

Alternate reality games like I Love Bees and Last Call Poker serving as promotional vehicles for traditional video games in the past. But few have tried to replicate Electronic Arts? ambitious attempt at fusing video game and transmedia experiences, Majestic. ?Sony returned to that concept?in 2009 with?PlayStation Xi, a console-based alternate reality game that created a game within the PlayStation Home virtual world.

This month, PlayStation returned to their familiar stomping grounds with a sequel,?Xi Continuum. Unlike its free-to-play precursor, players are asked to pay a nominal fee ($8.99) for the full Xi Continuum?experience. The return of?Xi?is one of many video game experiences that seek to directly interweave transmedia elements into the gameplay experience.?For Funcom?s The Secret World, PC-based gameplay is riddled with puzzle missions that require players to search for outside information to piece together clues scattered throughout the MMORPG?s world. These puzzle missions are punctuated by a series of alternate reality games that have?stretched back for years before the video game?s launch and continue today.

Alt-Minds looks to bring mission-based gameplay to a multi-lingual audience, offering versions of the game in French, English, German, and Spanish. The game frames the investigation around a web series. European players can turn to the game?s smartphone and tablet app to use augmented reality to hunt down additional clues at real world locations. Google is also making inroads into the augmented reality gaming market with its?Niantic Project alternate reality game. As a conspiracy-riddled narrative unfolds on the Niantic Project website, players across the globe battle to capture and maintain portals through the ARG?s augmented reality app, Ingress. Hacking portals in the augmented reality app periodically drops media files that advance the narrative, while codes and clues hidden within the narrative updates on the game?s website provide players with additional items and experience for the augmented reality game.

A Surge in Television and Movies

Syfy is looking to bridge the gap between video games and television with its ambitious upcoming project, Defiance. Syfy plans to have events in the online MMO shooter game influence the television series and vice versa, particularly as the endeavor, rumored to cost as much as $100 million, moves into its second season. Initial assets are beginning to proliferate online in anticipation of the show and game?s April premiere, offering a glimpse into the show?s post-apocalyptic universe.

Most transmedia endeavors in the realm of television and movies in 2012 were more modest in scope, seeking to offer curious viewers an amuse-bouche before the main event. For Cinemax?s show Hunted, the amuse-bouche took the form of a five-step job application to work at Byzantium Security, a company with the provocative slogan ?We?re not for everyone. Just the 1% that matters.? Sony created a similar teaser for its Skyfall campaign that challenged visitors to complete a text adventure as their application to MI6. Tim Kring tried a different tact with his fragmented web series?Daybreak 2012, which offered a thematic sequel to the show Touch that?explored the show?s mysterious ?doda? before the show?s second season. Investigation Discovery even created a condensed murder mystery?with a twist ending based on torrid love affair to promote?a reality series hosted by soap opera veteran Susan Lucci.

Even some of the larger movie-themed experiences of 2012 like the viral campaigns for?Prometheus, The Amazing Spider-Man, and The Dark Knight Rises focused on periodic teasers designed to make a big splash, like eccentric trillionaire Peter Weyland?s fictional TED Talk in 2023 for Prometheus and deaddrops containing Peter Parker?s school supplies and Spidey-shoes as part of Mark of the Spider-Man.

Social Samba served as the platform for a handful of the year?s more time-intensive extended television experiences, lead by USA Networks? Suits Recruits. The platform had previously been used for online extensions like Psych?s Hashtag Killer game and MTV?s Teen Wolf: The Hunt, but pushed the platform to new heights. Hearkening back to the Sherlock Holmes game 221B?s team-based gameplay, Suits Recruits required players to assume the role of either a paralegal or legal assistant at Pearson Hardman, share information with their compatriots to crack the case, and navigate the treacherous waters of office politics.

A Return to the Web Series

While traditional television and movies eased away from full-fledged alternate reality games in 2012, the web series community moved to fill the void. Leading the charge was Fourth Wall Studios with the official unveiling of the company?s RIDES platform through a dizzying array of shows, headlined by the Emmy Award-winning Dirty Work. As opposed to aiming for interactivity with the platform, it focused on adding additional layers of multimedia touchpoints to serve as a surround-sound of immersion. Stories ranged from the intensely dark science fiction show Flare to more lighthearted fare like the home shopping channel comedy RVC.

Guidestones was another platform-driven play aimed at rethinking the consumption of web video content: the story was designed to play out through a series of short videos emailed to subscribers at regular intervals, encouraging players to tackle the puzzles contained within in a fixed period of time. With a narrative centered around the very real Georgia Guidestones and a new season coming in 2013, the series matches its novel delivery system with an engaging story.

Tom Hanks also tested the transmedia waters through Tap Joint,?a perplexingly obscure alternate reality game that promoted the animated web series Electric City through a virtual telegraph machine that features prominently in the series. One of the game?s most innovative features was using the mobile version of the website as an instruction manual for the desktop version, a functional application of the narrative possibilities showcased in Randall Munroe?s April Fool?s joke on xkcd this year. Rounding out the big budget celebrity forays into the web series space,?Anthony Zuiker paired up with Yahoo and Norton Anti-Virus to produce?Cybergeddon, providing viewers of the chance to?team up with the show?s cyber-sleuths.

Some of the most engaging experiments in transmedia storytelling of 2012 came out of the established YouTube creator community. To support MyMusic, an over-the-top comedy about a radio station run by the physical manifestations of music genres, Benny and Rafi Fine started a series of weekly shows that have effectively brought the show?s fictional radio show to life. MyMusic serves as wish fulfillment for everyone who secretly fantasized about listening to the stations featured in WKRP in Cincinnati and NewsRadio. And then, there?s the Lizzie Bennet Diaries: a modern retelling of Pride and Prejudice spearheaded by Bernie Su and Hank Green that somehow manages to find endearing qualities in the the most unlikeable characters of the source material. It?s a rare Pride and Prejudice adaptation that keeps me coming back to find out what?s going on in the lives of ?Ricky? Collins and Lydia Bennet.

The Indie ARG Space

It?s getting increasingly difficult to draw the line between the indie space for alternate reality gaming and their better financed compatriots. Any one of the games I mention here (and many that I don?t) could easily claim I mislabeled them. Take, for instance, ZED.TO, a Toronto-based alternate reality game that held a series of elaborate live events that offer live theatrical experiences that rival many of the outrageous stunts pulled off during Tim Kring?s Conspiracy for Good. Similarly, the quality of the radio drama featured in the We Are the Earthborne ARG leaves me nostalgic for the halcyon days of audio dramas. I can?t even begin to make sense of TVTropes.org?s recently concluded alternate reality game, The Wall Will Fall, but its almost perverse complexity riddled with meta-references and in-jokes seems perfectly tailored to the community?s near-herculean organizational prowess. Alternet Reality delves into the horror genre through intense, live streaming challenges where the fictional characters? fates depends on players? ability to crack the case in real time.

The independent ARG that stuck with me the most this year, however, made no attempt to create the illusion of being anything but an indie project. On a lark, ARGNet founder Steve Peters announced an insta-ARG experiment parody of World Without Oil dubbed?World Without Helium. The Mega Hard Wood Group emerged from that framework, exploring the serious topic of a world facing a helium shortage in the most whimsical manner possible. The game?s protagonist William Sawtooth III decided that if corporations can be people, people should be able to function as corporations as well. So Sawtooth sold shares in his personhood, putting his life at the whims of his shareholders. The game only ran for two weeks, but every day brought a new relatively simple yet ridiculous challenge delivered through a podcast to recover William Sawtooth?s shares from the Mega Wood Group board of directors that still managed to emerge organically from the story. Other games like Work With No Pants?also embraced the absurdist aesthetic to great effect.

Persistence of the Serious Game

One of the most powerful serious games of 2011 returned to the USC campus this fall. Reality Ends Here was designed as a way of encouraging USC?s freshman film students to continue the practical experimentation and creative spark that got students admitted to the School of Cinematic Arts in the first place outside the classroom context. The project?s inaugural year won Indiecade?s Impact Award. Reality Ends Here returned to meet a new crop of incoming students this fall. The year?s projects included everything from a massive science fiction supercut remix, vignettes from an Israeli soldier?s struggles in the military, and a film created using Google Maps in place of a camera.

Education was a popular topic for serious ARGs this year, with?World Without Oil creator Ken Eklund returned to the space for?Ed Zed Omega, a game about high school dropouts with a twist. Instead of using Ed Zed Omega to encourage students to work with the current educational system, the game follows six fictional teens to address what steps high school drop-outs take when they make that decision.?ARGs were even used to promote attending Easter services for Long Hollow Baptist Church, through the campaign Who is Phillip Randoll. A light puzzle-driven narrative was used alongside out of home advertisements to build up anticipation for church?s Easter drama, The Fortunate Death of Phillip Randoll.

Books and Puzzle Challenges

For the book publishing industry, 2012 was dominated by a continuation of existing projects. 39 Clues continues to push through its second series, Yomi Ayeni?s Clockwork Watch steampunk graphic novel shipped,?and the upcoming transmedia series The Karada has been teasing its eventual release with the Wattpad story All Your Fates. Eli Horowitz, author of the recently solved puzzle book The Clock Without a Face, recently released The Silent History, a book designed exclusively for iDevices that can unlock location-specific stories for its readers. For non-fiction resources, 2012 saw the publication of Andrea Phillips??Creator?s Guide to Transmedia Storytelling?and Tyler Weaver?s?Comics for Film, Games, and Animation.?There are also a number of transmedia book experiences set to come out in 2013,?including Thomas Greanias thriller The Alignment: Federal City and Mark Staufer?s lucid dreaming exploration The Numinous Place.

Oddly enough, some of the most exciting news items relating to the publishing world are tied to already published books. At ARGFest this year, it was announced that there are four meta puzzles hidden in Dave Szulborski?s book, This Is Not A Game?that have remained hidden within the book for over seven years. Ernest Cline also revealed that his book?Ready Player One contained meta-puzzles that mirrored the novel?s plot, and offered up a 1981 Delorean as the prize for the first to complete a series of three increasingly difficult challenges. First, competitors needed to find a secret room in an emulated Atari game created for the challenge. Next, puzzlers had to collect 80?s-themed items in Richard Garrett?s Facebook game, Ultimate Collector. Finally, everyone to clear the first two challenges were asked to set a new world record on one of six classic video games. Within eight days, the contest?s winner set a new world record on Joust, and Cline gave the Delorean away on G4TV.

A handful of other major puzzles hunts concluded their multi-year runs. For?The Clock Without a Face, the last of twelve clock faces was located after players spent two years scouring the book for clues. Hints at major campaigns for 2013 are already beginning to surface. JJ Abrams? new?Star Trek film hid a sign-up page for a new viral experience in the film?s newest trailer, and Google?s?Niantic Project game is still in closed beta, with prospective players chomping at the bit to start competing for world domination.?Animism has returned for a second alternate reality game launching on January 1st with Grind City University, there?s bound to be something for everyone.

I inevitably neglected to mention more than a few key moments of the year, like?Homestuck?s Adventure Game raising almost $2.5 million on Kickstarter, an international art heist game that culminated with an event at the European Parliament, or my unrepentant glee at finally getting to hear Jack and Eugene bicker as I play Zombies, Run.?Feel free to share your own favorite moments in the comments, or check out our look back at the highlights from 2011.

Live and learn: Everybody went mobile in 2012 (or so it seemed), but the most groundbreaking movement on the Web may have been the rise of digital education.

For all the attention lavished on the Web?s growth on mobile devices this year, one of the most interesting Internet trends is still best experienced on a desktop computer: online education.

The rising cost of higher education (the average bachelor?s degree now costs more than $100,000), combined with increasing access to high-speed Internet service and a desire for more efficient and flexible learning methods, brought new prominence to websites offering free or low-priced courses in everything from programming to literature.

Free online code-learning startup Codecademy?s effort to teach novices to code snagged more than 400,000 participants for its weekly lessons in JavaScript, HTML, and CSS. Harvard and MIT joined forces to create edX, a $60 million nonprofit company that streams free college courses online, while nearly three dozen schools?including Stanford and Princeton?formed their own free online course site, Coursera, which has more than 1.5 million users so far.

Udacity, cofounded by Sebastian Thrun, a Google Fellow and former Stanford researcher, started out by offering a single Stanford artificial intelligence class online for free. It has since grown and now offers 19 different free courses, mostly geared toward computer science and math. And Duolingo, a free crowdsourced language-learning startup cocreated by Carnegie Mellon University professor Luis von Ahn, has about 300,000 users per week learning French, Spanish, English, Italian, German, and Portuguese.

Perhaps the most ambitious (and highly funded) online education offering unveiled in the last year was the Minerva Project, which raised $25 million from Benchmark Capital for its plan to offer a completely online college education for about $25,000 a year. We?ll have to wait to gauge the Minerva Project?s impact, though: it?s not starting classes until 2015.

Despite the initial wave of enthusiasm, it?s not yet clear whether many of these startups or universities will be able to form sustainable business models, or if online classes can really work well on a large scale (many of the students that sign up for classes don?t actually complete them).

Fortunately, because they operate on the Web, these education efforts are able to gather lots of data about how their students are learning?potentially useful for tweaking lessons and improving performance.

Another segment of the Web that experienced major changes in 2012 was social networking. More than eight years after it began in Mark Zuckerberg?s Harvard dorm room, Facebook crossed the billion-user milestone in October.

The leading social network also became a publicly traded company, albeit not a particularly successful one; the stock began trading on May 18 at $42; as of Wednesday, it had declined to $26.51. This move means that Facebook, which is constantly gathering more data about our lives, is now facing even greater pressure to find ways to profit from this information. Facebook bought the popular photo-sharing service Instagram in a bid to beef up its own photo-sharing offerings and capitalize on Instagram?s influence as a standalone app. Facebook set the acquisition price at $1 billion in April, but the stock and cash deal was worth about $715 million when it closed in August due to the drop in Facebook?s share price.

Pinterest, a social-curation site founded by Ben Silbermann that lets you ?pin? interesting items found online to virtual pinboards, emerged in 2012 as one of the most popular social networking sites. Though Pinterest launched back in 2010, it suddenly ascended early this year. In April, a report from Experian Marketing Services named it the third most popular social network behind Facebook and Twitter.

Of course, more and more of the Web was experienced on smartphones and tablets in 2012. The first time smartphones and tablets outsold personal computers?was 2011, and that trend continued in 2012. Mobile Internet usage has been rising quickly?it accounted for 13 percent of Web traffic in November?and it?s expected to grow more dramatically. According to IDC, the number of mobile users is expected to climb from 174 million this year to 265 million in 2016, with more of us accessing the Web with a mobile device than with a PC in 2015. Staphylococcus aureus: Why it just gets up your nose

Dec. 27, 2012 ? A collaboration between researchers at the School of Biochemistry and Immunology and the Department of Microbiology at Trinity College Dublin has identified a mechanism by which the bacterium Staphylococcus aureus (S. aureus) colonizes our nasal passages. The study, published December 28 in the open access journal PLOS Pathogens, shows for the first time that a protein located on the bacterial surface called clumping factor B (ClfB) has high affinity for the skin protein loricrin.

S. aureus is a major human pathogen, with the potential to cause severe invasive diseases. It is a major cause for concern in hospitals and healthcare facilities, where many infections are caused by strains resistant to commonly used antibiotics [MRSA]. Interestingly, S. aureus persistently colonizes about 20% of the human population by binding to skin-like cells within the nasal cavity. Being colonized predisposes an individual towards becoming infected so it is vital that we understand the mechanisms involved.

ClfB was previously shown to promote S. aureus colonization in a human nasal colonization volunteer study. This paper now identifies the mechanism by which ClfB facilitates S. aureus nasal colonization. Purified ClfB bound loricrin with high affinity and this interaction was shown to be crucial for successful colonization of the nose in a mouse model. A knockout mouse lacking loricrin in its skin cells allowed fewer bacterial cells to colonize its nasal passages than a normal mouse. When S. aureus strains that lacked ClfB were used nasal colonization could not be achieved at all. Finally it was shown that soluble loricrin could reduce binding of S. aureus to human nasal skin cells and that nasal administration of loricrin reduced S. aureus colonization of mice.

Rachel McLoughlin, the study's corresponding author and Lecturer at the School of Biochemistry and Immunology at Trinity College Dublin concludes, "Loricrin is a major determinant of S. aureus nasal colonization." This discovery therefore opens new avenues for developing therapeutic strategies to reduce the burden of nasal carriage and consequently infections with this bacterium. This is particularly important given the difficulties associated with treating MRSA infections.

In the first week of 2000, local New York City hospitals recorded an astonishing 50.8 percent more deaths than in the last week of 1999, according to the New York Times. Apparently, a significant number of people delayed their deaths in order to see the new millennium.

In the summer of 2004 something very strange happened in Australia. The birthrate plummeted sharply in June. Then on a single day in July more babies were born than on any other day in the prior thirty years of Australian history. July 1, 2004 was a very popular day to be born.

What caused this dip and surge in births? Seven weeks before July 1, the Australian government announced a change in the tax code that would give families a $3,000 baby-bonus starting on the first day in July. It appears that as many as 1,000 births were "moved" until after the baby bonus kicked-in, according to a 2009 study by Joshua Gans of the University of Melbourne's Business School and Andrew Leigh of the Australian National University's Research School of Social Sciences.

"We estimate that over 1,000 births were "moved" so as to ensure that their parents were eligible for the Baby Bonus, with about one quarter being moved by more than one week," Gans and Leigh write.

According to the researchers, most of this temporal shift was due to changes in the timing of induction and cesarean section procedures. When they looked at an increase in the baby bonus that took effect two years later, on July 1, 2006, they found that the same pattern?births moving from June to July.

Now, some of this could be due to fudging the dates, so that births occurring just prior to the eligibility date were recorded as taking place afterward. But Gans and Leigh found evidence that babies really were held-in-uteri until they were eligible for the tax break. Babies born after the eligibility date, for example, had higher birth weights than those born earlier.

An earlier paper by Gans and Leigh looked into another natural experiment. In 1979, Australia abolished its federal inheritance taxes. Official records show that approximately 50 deaths were shifted from the week before the abolition to the week after.

"Although we cannot rule out the possibility that our results are driven by misreporting, our results imply that over the very short run, the death rate may be highly elastic with respect to the inheritance tax rate," Gans and Leigh write.

This isn't just something peculiar to Australia. Economists Wojciech Kopczuk of Columbia University and Joel Slemrod of the University of Michigan studied how mortality rates in the United States were changed by falling estate taxes. They note that while the evidence of "death elasticity" is "not overwhelming," every $10,000 in available tax savings increases the chance of dying in the low-tax period by 1.6 percent. This is true both when taxes are falling, so that people are surviving longer to achieve the tax savings, and when they are rising, so that people are dying earlier, according to Kopczuk and Slemrod.

"Death elasticity" does not necessarily mean that greedy relatives are pulling the plug on the dying or forcing the sickly to extend their lives into a lower taxed period. According to a 2008 paper from University of Pittsburgh Medical Center Doctor G. Stuart Mendenhall, while tax increases give potential heirs large economic incentives to limit care that would prolong life, distressed patients may "voluntarily trade prolongation of their life past the end [a low tax period] for large ?nancial implications for their kin.

"Whether these incentives are explicitly speci?ed in wills or communicated to their power of attorney over the dinner table, they are clearly present and affect the ability of all involved parties to make unbiased decisions," Mendenhall writes.

We had something of a natural experiment in death and taxes in 2010, when the estate tax was eliminated for one year. Many predicted that this would result in many fewer deaths at the end of 2009 and a surge in deaths prior to taxes rising in 2011.

My own research hasn't uncovered any formal academic work on this period. Perhaps it is too recent. Or perhaps the setting of the exemption at $5 million made the sample size of those that could achieve significant tax savings by dying in 2010 rather than 2011 too small.

But based on past reactions to changes in taxes, it at least seems likely that some deaths that might otherwise have occurred shortly after January 1 will occur shortly before. Death may slip in ahead of the tax man for some with estates worth over $1 million.

The business community has been pretty quiet in the wake of the horrific shootings at Sandy Hook. But I have found some of the most interesting and compelling journalism in the aftermath to be focused on the firearms industry, itself, and how it is regulated (or isn't). Five I've found the most thought-provoking:

1. NPR's report on why the AR-15, the weapon used in the massacre, has become so popular. One explanation: as the number of gun enthusiasts wane, gun manufacturers are marketing more expensive (and more lethal) models to a shrinking pool of customers.

This article is primarily directed toward people who work at home in a business that happens to involve teamwork in a network marketing environment.

I am often contacted by members of my business team with complaints regarding a lack of response or lack of action on the part of their affiliates. Their complaints typically go something likeI send out lots of emails to all of them and I very rarely get any response and it seems like very few of them are taking any action. What am I doing wrong?.

There are many types of internet-based home businesses where feedback and interaction are quite important and getting an affiliate or business team member to communicate with you is a definite objective. Its not at all surprising that this can be a challenge when you stop to think that what you are trying to do is initiate a dialog between two complete strangers who have never even seen each other.

In the case of network marketing it is of critical importance to build a sense of trust with your team members. Network marketing is very definitely a relationship business and each relationship typically starts out with two complete strangers sitting at their respective keyboards.

The team building aspect of network marketing is of vital importance to the success of each individual member and to the team as a whole. Two key elements of team building are trust and confidence and these factors play a very large role particularly for new team members.
